Papst Franziskus X sitzt in einem Dreiviertel-Profil, wie er nach links blickt. Er wird von zwei Verwandten flankiert, Kardinal Giulio de’ Medici links und Kardinal Luigi de’ Rossi rechts. Leo X hält eine Lupe in der Nähe der beleuchteten Bibel und Glocke auf dem Tisch. Der Pommel auf dem Stuhl des Papstes ruft die Kugeln des Medici Wappens hervor. Andrea del Sarto kopierte dieses Gemälde von Raphaels Original (1518-19) auf die Kommission von Ottaviano de’ Medici. Auf Ersuchen des Papstes Clement VII (gekennzeichnet als Kardinal Giulio de’ Medici in der Malerei), Ottaviano wurde gebeten, das Portrait an Herzog Federico Gonzaga in Mantua zu senden. Ottaviano schickte Andrea del Sartos Kopie nicht mit Raphaels Original, sondern ohne Nachweis. Die Details dieser Veranstaltung werden in Giorgio Vasaris berühmtem Leben der Künstler aufgenommen (veröffentlicht 1550, 1568).
