Ippolito dei Medici (1511-1535) était le fils naturel de Giuliano, duc de Nemours. Alors qu'il était encore bébé, il fut confié aux soins de son oncle, le pape Léon X (Giovanni Medici). Le cousin du pape, qui lui succéda de devenir le pape Clément VII, fit d'Ippolito un cardinal en 1529. Sa tenue inhabituelle est une référence à la partie qu'il a jouée dans la défense de la Hongrie contre les Turcs, peu avant la création de ce portrait.
