Vasco da Gama nacque a Sines, probabilmente nel 1468, e morì in India nel dicembre 1524, solo tre mesi nel suo mandato come viceré dell'India portoghese. Era il secondo figlio di Estêvão da Gama. Suo fratello maggiore era Paulo da Gama, che viaggiò con Vasco durante il viaggio in India del 1497-1499 e morì quasi alla fine del viaggio di ritorno all'isola di Terceira nelle Azzorre.
