Il famoso episodio di Susanna, che nella pittura veneta era stato ripreso da Jacopo Tintoretto con alcuni dipinti intorno al 1560, è narrato nel Libro di Daniele. La giovane donna, colta di sorpresa durante un bagno da due uomini, respinse i loro progressi e, ingiustamente accusata, fu infine salvata da un intervento provvidenziale di Daniel. La grande capacità di narrazione di Palma emerge chiaramente in questa tela. Spiega l'episodio su vari piani receding, concentrandosi sul nudo centrale curvo di Susanna sostenuto da una cameriera, mentre i due anziani si nascondono dietro un albero—un dettaglio non privo di un certo umorismo.
