San Girolamo (ca. 342–420), uno dei quattro grandi dottori della Chiesa occidentale, è venerato per la sua pietà ascetica e per la sua monumentale traduzione latina della Bibbia, rappresentata qui dal grande volume su cui poggia le mani. A seguito di una vecchia convenzione, l'artista lo raffigura nelle vesti di un cardinale. Questa composizione si è rivelata popolare ed è stata prodotta in almeno quattro versioni da El Greco e dal suo negozio; una di queste è nel Metropolitan Museum of Art di New York City.
