San Girolamo (circa 342 - 420), l'ispirazione principale per i penitenti e gli eremiti cristiani, è mostrata a sinistra con un crocifisso. San Nicola di Tolentino (circa 1245 - 1305) indossa l'abitudine nera dell'ordine agostiniano e tiene gigli sul suo seno, che brilla di splendore sacro. La bara su cui Cristo sta è più insolita e può essere presa per riferirsi alla sua futura conquista della morte. Questa opera fu dipinta verso la fine del periodo che Lotto trascorse principalmente a Bergamo. C'è un'altra, meglio conservata, versione del quadro (apparentemente dipinto allo stesso tempo ma disegnato prima), nel Museo di Belle Arti, Boston.
