Le tre teste alludono alle tre età dell'uomo: gioventù, maturità e vecchiaia. L'iscrizione è articolata in tre sezioni associate alle rispettive teste sottostanti. La testa sinistra assomiglia a Tiziano stesso in età avanzata; l'uomo centrale barbuto è stato pensato per rappresentare suo figlio Orazio, mentre i giovani possono raffigurare suo cugino ed erede, Marco Vecellio (nato 1545). La bestia a tripla testa - lupo, leone e cane - è un simbolo di prudenza.
